The group of Dr Ghisleni (Mechano-Oncology Lab at IFOM, IFOM-IEO campus, Via Adamello 16, Milano) is looking for a highly motivated and dedicated master student interested in in vitro models of cardiovascular fibrosis progression. The project focuses on the development of physio mimetic model for culturing primary cardiac and pulmonary fibroblasts as a tool to study the biochemical and mechanical pathways that activate the fibrotic response. Dr. Ghisleni has a background in structural biology with deep knowledge in mechanobiology, with a focus on novel cell biophysical and fluorescence imaging techniques. The student will be trained in the isolation and characterisation of murine primary cells.
S/he will learn
(I) the culture of mammalian primary cells in 2D and 3D physio-mimetic substrates,
(II) the characterisation of phenotypic plasticity,
(III) immunofluorescence and imaging (live-cell and confocal microscopy) for visualization and quantification of biomechanical parameters.
The aims of this project will be the following:
(I) Isolate and cultivate primary murine fibroblasts from cardiac and lung tissues.
(II) Mechanobiology approaches to engineer the composition and mechanical properties of culturing substrates.
(III) Biophysical investigations of cell biomechanical properties in the context of Fibroblast-to-Myofibroblast phenotypic plasticity.
